Tugas Akhir EKSPRESI PROTEIN P53 DAN CYCLINE DEPENDENT KINASE (CDK) PADA KANKER MAMMAE MENCIT SETELAH PEMBERIAN FRAKSI ALKALOID JARONG (ACHYRANTHES ASPERA LINN)
Abstrak EXPRESSION OF P53 PROTEIN AND CYCLINE DEPENDENT KINASE (CDK) ON MICE BREAST CANCER GIVEN ALKALOID FRACTION JARONG (ACHYRANTHES ASPERA LINN) I DEWA PUTU ANOM ADNYANA Empirically and scientifically Achyranthes aspera linn leaf is used to treat cancer, particularly breast cancer and cancer cervic. Scientifically proven alkaloid fraction of leaves Achyranthes aspera linn cause of death in accordance with the mechanism of myeloma cell apoptosis and necrosis associated with p53 gene and protein suppresor cycline dependent kinase enzymes. This research used animal experiment female mice aged 2 months were 60 tails that were made to suffer mammary cancer by benzo(α)pyrene with a dose of 10 mg/kgbw subcutaneously mammary area for eight weeks with injections of 3 days interval. Occurrence of mammary cancer in mice proved to have a lump on the macroscopic and microscopic mammary via dissection of mammary cells that have experienced proliferation. Mice were divided into six groups so that each group consisted of l0 rats. Groups: negative control group, positive control group of cyclophosphamide 6 mg/kgbw, the treatment group PI, P2, P3 and P4 are given alkaloid Achyranthes aspera linn continuous dose of Na 0.5% CMC (0 mg/kgbw), 10 mg/kgbw; 30 mg/kgbw and 100 mg/kgbw. All groups received treatment every third day for eight weeks. The parameters were observed with immunohistochemistry to see the amount of p53 protein expression and total expression Cycline Dependent Kinases (CDK). The results of this study Achyranthes aspera linn alkaloids caused an increase in the amount of p53 protein expression and a decreased in the number of significant expression of CDK enzymes between control and treatment groups. The conclusion was the alkaloid fraction of leaves of Achyranthes aspera Linn at a dose of 100 mg / kg body weight given to mice suffering from mammary cancer could increase NK cells, apoptotic bodies cell, necrosis cell, p53 gene protein and lower the viability, and CDK enzymes. In the toxicity test of the alkaloid fraction of leaves of Achyranthes aspera Linn was a very safe material used in medicine.
