THE ANALYSIS OF HEPATITIS B VIRUS (HBV) GENOTYPES, SUBGENOTYPES AND SUBTYPES ON S (Surface) REGION GENES
FROM PATIENT WITH HBsAg POSITIVE
IN MENGWI, BADUNG, BALI
Indonesia has a high endemic level of hepatitis B infection. Since the last decade, Mengwi has become a developed region with rapid grow of social-ecomomics, demography and dynamic inhabitant . This condition can emerge variations of hepatitis B virus (HBV) genotype, subgenotype and subtype. The purpose of this research is to analize the HBV genotype, subgenotype and subtype on patient with hepatitis signs and symptoms who comes to Puskesmas Mengwi I, Badung, Bali.
All serum samples were examinated for ALT/SGPT level and detection of HBsAg. Serum of HBsAg samples positive were used for identification of HBV genotype, subgenotype and subtype. We carried out DNA extraction, amplification with polymerase chain reaction and sequencing of serum samples. The obtained nucleotide sequences were compared from international DNA data bank for HBV genotypes and subgenotypes determination. HBV subtype were determined using analysis of amino acid substitutions at positions 122, 127, 134, 159, 160, and 177 of S gene.
This study used 75 serum samples that taken from patients. Two patients (2,7%) were found with high level of ALT. HBsAg positive were found in 11 samples (14,7%). HBV genotype B were found predominantly, ten of eleven isolates belonged to genotype B and one to genotype C. Seven examined isolates belonged to subgenotype B3, three to subgenotype B7 and two were not classified to any of subgenotype B yet because they did not fit in any branch of subgenotype B on the phylogenetic tree. Subtype adw2 was found on ten isolates (90.9%), followed by subtype adrq+ (9.1%). Interestingly, all of genotype B were subtype adw2, and all of genotype C were subtype adrq+ .
